《乐一通秀》将兔八哥和达菲鸭设定为一对住在一起的室友，他们生活在现代郊区，与猪小弟、崔弟、傻大猫、大嘴怪等熟悉的角色成为邻居或朋友。每集围绕他们的日常琐事展开，例如兔八哥试图维持生计、达菲鸭的各种异想天开的计划，以及他们与周边人物的有趣互动。节目中还穿插了“大头针”音乐视频和“快速鸟与大灰狼”动画短片，为观众带来更多经典元素。
